Blood-based microRNA liver biomarkers – hepatomiR®

The hepatomiR® kit (CE-IVD) is intended for the quantification of hsa-miR-122-5p, hsa-miR-192-5p, and hsa-miR-151a-5p in human plasma samples for diagnostic use.

A software application included with the kit computes a liver function score (hepatomiR® P-Score) using a proprietary algorithm.

The hepatomiR® P-Score has been proven useful to assess the risk of post hepatectomy liver failure (PHLF) in patients with HCC, CCC, or metastatic colorectal cancer (Starlinger et al., HEPATOLOGY, Vol. 69, No. 6, 2019). This is critical to identify patients with a high risk of postoperative liver dysfunction and to plan appropriate risk reduction measures.

Other currently investigated intended uses of the product are the diagnosis acute drug-induced liver injury and the diagnosis and monitoring of metabolic associated liver disease (MAFLD).

One kit provides sufficient reagents for the analysis of 48 samples.

The hepatomiR® kit is intended to be used with human samples. However, orthologs for all hepatomiRs exist in other species, enabling the use of the tests in non-mammalian species for research-use purposes. Importantly, the hepatomiR P-score has not been validated for non-human samples. To assess the feasibility of such projects, please contact us before using the assay in non-human species.

We have performed an in-depth analysis of the correlations of hepatomiR® P-score to parameters such as liver enzymes and ICG clearance. Read our key publication to find out how hepatomiR® performs in comparison to these parameters.

The hepatomiR® kit (CE-IVD) is compatible with Roche LC480 I and II instruments and supports 96-well and 384-well plate formats.

Three different quality control assays for monitoring analytical performance (RNA isolation, reverse transcription, and PCR) as well as two quality controls to assess sample quality (hemolysis) are included in the kit. No further controls are necessary to run an analysis. The data analysis tool automatically performs a QC pass/non-pass check for each analyzed sample.

The hepatomiR® kit requires the use of double-centrifuged plasma in order to obtain reliable results. Please refer to page 21 of this manual for the specific collection protocol. Important: Heparin as an anticoagulant is not compatible with RT-qPCR analysis and must be avoided. CTAD (Citrate, Theophyllin, Adenosin and Dipyridamol) has been exclusively used during the development of the hepatomiR® kit. Citrate has been shown to yield comparative results.

The hepatomiR® kit includes the Plasma RNA extraction kit for RNA isolation protocol from a defined input volume of 200 µL plasma. The kit ensures high RNA yield and prevents inhibition of downstream PCR amplification. For further information on RNA isolation please refer to the sections in the instructions-for-use.

The intra-assay precision reflects variability among replicate determinations of the hepatomiR p-score within the same assay run. For this experiment, three donors were measured in quadruplicates on one day within one analysis run. The intra-assay standard deviation (SD) of the hepatomiR P-Score for the three donors was found to be between 0.05 and 0.01.

Inter-assay precision measures the variance between runs of sample replicates on different plates and can be used to assess plate-to-plate consistency. HepatomiR p-score analysis workflow was performed on three different days.  An inter-assay SD of the hepatomiR P-Score between 0.04 and 0.02 was determined.
